$AMRSQ I wanted to outline another feature of the three COVID-19 trials discussed by PSS. In the THEMBA trial #2, the plan is to assess a protein-based vaccine + adjuvant. The original study uses the 3M-052-alum adjuvant (see title below). However, a subsequent COVID-19-related study uses the same 3M-052 adjuvant, but this time formulated with squalene (not alum): 3M-052-SE. While the studies involve different institutions, IDRI was involved with both - in particular, a single author Christopher B. Fox, with IDRI specifically acknowledge for 3M-052-SE formulation. Given that PSS wants to scale these vaccine platforms, this second study demonstrates that the 3M-052 adjuvant + squalene is effective and likely paves the way for the THEMBA trial #2 to switch from 3M-052-Alum to 3M-052-SE (squalene). In other words, my expectation is that Amyris squalene will be incorporated into both the THEMBA trial #1 and #2 trials.

Among the demonstrated benefits of squalene emulsions is the ability to enable vaccine antigen dose sparing, an important consideration for pandemic response. In order to increase pandemic response capabilities, it is desirable to scale up adjuvant manufacturing processes.
